DIM Detox is Pure Encapsulations' most comprehensive hepatic detoxification and hormone metabolism support formula—a ten-ingredient combination that addresses oestrogen metabolism, phase II liver detoxification, antioxidant protection, and support for the glucuronidation pathway in a single, clinically formulated product. Unlike simpler DIM-only supplements, this formula is designed around the understanding that healthy oestrogen processing requires not just the initial metabolic steps but the full downstream detoxification cascade — from phase I oestrogen hydroxylation through phase II conjugation and elimination.
The formula operates across four complementary functional mechanisms:
1. Oestrogen Metabolism Support — BioResponse DIM® and Broccoli Sprout Concentrate
BioResponse DIM® (Diindolylmethane Complex) — 100mg (providing 25mg DIM) Diindolylmethane (DIM) is a stable metabolite of indole-3-carbinol (I3C), naturally formed when cruciferous vegetables are chewed and digested. DIM's principal mechanism is the promotion of favourable oestrogen metabolism — specifically, it supports the 2-hydroxylation pathway of oestrogen metabolism, favouring the production of 2-hydroxyoestrone (2-OHE1) over the more biologically active and potentially proliferative 16α-hydroxyoestrone (16α-OHE1) and 4-hydroxyoestrone (4-OHE1). This shift in the 2:16 oestrogen metabolite ratio is considered a meaningful marker of healthy oestrogen metabolism in both women and men.
Plain DIM has poor water solubility and bioavailability. BioResponse DIM® is a patented delivery system (US Patent 6,086,915) that complexes DIM with starch, phosphatidylcholine, vitamin E succinate, and silica — creating a microencapsulated complex that significantly enhances DIM's absorption and oral bioavailability compared to unformulated DIM powder. The 100mg complex provides 25mg of actual DIM per serving.
Broccoli Sprout Concentrate (Brassica oleracea italica, whole plant, standardised to minimum of 400 mcg sulforaphane) — 100mg Sulforaphane, the principal bioactive derived from glucoraphanin in broccoli sprouts, is the most potent known inducer of the Nrf2 (nuclear factor erythroid 2-related factor 2) transcription pathway — the master regulator of the body's endogenous antioxidant and phase II detoxification enzyme systems. Calcium D-Glucarate™ (patented, Applied Food Sciences) — 100mg As covered in the standalone Calcium D-Glucarate listing, this ingredient supports healthy hormone elimination through the glucuronidation detoxification pathway by inhibiting beta-glucuronidase — the enzyme that can deconjugate and reactivate oestrogen metabolites before they are excreted, allowing them to re-enter circulation. By supporting beta-glucuronidase regulation, calcium D-glucarate helps ensure that conjugated oestrogen metabolites complete their elimination cycle rather than being reabsorbed. This mechanism is directly complementary to DIM's role in promoting favourable oestrogen hydroxylation — DIM shifts which oestrogen metabolites are produced, while calcium D-glucarate supports their efficient elimination.
3. Phase II Liver Detoxification Support — Milk Thistle, Alpha Lipoic Acid, NAC, and Amino Acids
Milk Thistle Extract (Silybum marianum, seed, standardised to 80% silymarin) — 150mg Silymarin — the flavonolignan complex from milk thistle seed — is one of the most extensively researched hepatoprotective botanicals. It supports phase II detoxification enzyme activity, provides antioxidant protection to hepatocytes, promotes hepatic glutathione levels, and has demonstrated liver membrane-stabilising properties. The 80% silymarin standardisation ensures consistent delivery of the active fractions responsible for these effects.
Alpha Lipoic Acid (ALA, thioctic acid) — 200mg ALA is a universal antioxidant — unique in its ability to quench both fat-soluble and water-soluble free radicals — and a critical cofactor for mitochondrial energy metabolism. In the context of liver detoxification, ALA supports phase II enzyme activity, regenerates endogenous antioxidants including glutathione and vitamins C and E, and provides direct antioxidant protection during the oxidative burden associated with phase I detoxification reactions.
N-Acetyl-L-Cysteine (NAC, free-form) — 200mg NAC is the immediate precursor to L-cysteine, the rate-limiting substrate for glutathione synthesis. 4. Lignan Support — HMRlignan™
HMRlignan™ (Norway spruce knot wood, 7-hydroxymatairesinol) — 10mg HMRlignan™ is a patented lignan extract from Norway spruce (Linnea Inc.) providing 7-hydroxymatairesinol (HMR), a plant lignan converted by intestinal bacteria to the mammalian lignan enterolactone. Enterolactone has demonstrated weak oestrogenic and anti-oestrogenic properties depending on cellular context, and research indicates that higher dietary lignan intake and healthy serum enterolactone levels are associated with supportive outcomes for cellular, cardiovascular, bone, and cognitive health.
